A gain-of-function (GOF) model's genetic support is equally compatible with a loss-of-function (LOF) framework. This stems from the tendency of proteins rendered unstable by mutations (such as APP in Alzheimer's disease, or SNCA in Parkinson's disease) to aggregate and become depleted from the soluble protein pool.
